No, they’re not in the same universe, as Rockstar confirmed.
I'd presume that the events of Red Dead Revolver happened in the same universe, but maybe a bit differently, since yeah, they aren't actually in the same universe, but Red Harlow has been mentioned and you can play as him in the RD Redemption Multiplayer (which however i don't think is canon either)
Gonna have to agree with Wag. Dan Houser mentioned in a GameSpot interview regarding undead nightmare that revolver is in its own canon.

Yes, they all take place within the same universe. You never see any of the characters from Red Dead Revolver (I don't think you do at least), but I know that people will sometimes tell stories about Red Harlow when sitting around a campfire
Red Dead Revolver isn't canon.
However, it is still implied that some of the Revolver characters do exist in the Redemption universe as folk legends that may or may not have existed.

The story of Red Dead Revolver is 100% a folktale within the Redemption universe.
It’s not a true story but the story it self does exist.
I don't know if Revolver's story is "100% folklore".
I went back to record some of the campfire tales and some of them talk about the Revolver characters and places as if they exist.
For example, the Mexican people disliked General Diego and wanted him dead, which would be weird if he was just a fictional character.
The characters talk about Brimstone as if it was a real place. One story even implies that the Brimstone quick draw competition went defunct.
^Have you ever heard of the unreliable narrator? This could be an example of that.
Meaning the characters telling you a story could have been written to be incorrect about what they’re saying. This happens in The Elder Scrolls a lot.
The places and maybe some of the characters could possibly exist but the stories are still just legends in my opinion. The stories probably take place in real locations though.
This sort of thing happens in real life.
For example King Arthur was never a real person but I guarantee some people think he was because in his stories he ruled over a real place.
The Trojan Wars and the Greek’s invading a city by hiding in a giant horse never really happened. I know a lot of people think thats a real story too.
I mean, the King Arthur and the Trojan Horse stories had historical basis. King Arthur never existed, but he was based on multiple figures. There's evidence that the Trojan War happened, though definitely not as grand and epic as the Iliad made it out to be.
Plus, I think the timeline is way too close for fictional characters to be confused with real ones. The events of Revolver happen sometime in the mid-1880's. I don't find it likely that the Mexican people highly disliked a man that never existed.
I think the community is taking all of these Red Dead Revolver references too seriously. The campfire stories about Red Harlow are more or less Easter eggs. I wouldn’t presume that Rockstar put them in as serious, canonical fact.
Dan Houser and Rockstar has said multiple times that Revolver and Redemption are two different universes. They’ve been pretty clear about it.
