From a location standpoint, it's a bit of a toss up between Horseshoe Outlook and Clemen's Point. Clemen's edges out slightly, due to being lake side.
As far as content, you really need to get into chapter 3, unfortunately, to unlock some really good content.

I read a part that said they had scoped out safehouses in New Austin, for after the ferry job.

I'd like a DLC where Arthur can be in New Austin, without glitching. I don't have anything against John, but i don't have any interest in exploring New Austin, as him, in RDR 2.
I like Clements Point for the easy access to resources (water, fish and hunting) but i enjoyed the eerie atmosphere at Shady Belle, as well.
Most cosmetic things, from online, should be available in SP. Outside of that, i can't think of much more. They did a pretty good job of fleshing out the side activities, this time.
